What are Peptide Pools?

Peptide pools are clusters of immunostimulatory antigens, which are large collections of small peptides of specific lengths and different sequences. It consists of arrangements and combinations of various (or most) amino acid sequences in short peptides of that length, and is suitable for T cell stimulation of HIV, HCV, influenza, and other infectious diseases. It can be used for drug design, protein-protein interactions, biochemistry and pharmaceutical applications. Peptide pools are built based on small peptides containing key residues (mimic antigenic determinants clusters on proteins) as well as protein-protein interactions and recognition (interactions between local peptide fragments).

First, the desired peptide sequences need to be designed according to the purpose of the experiment. These sequences can be linear, cyclic, or contain modifying groups. Next, amino acid residues are added stepwise by solid-phase synthesis and peptide chains are formed by activation, coupling and deprotection. The prepared peptide chains are then collected together, either by physical mixing or by using branching protection during the synthesis process, resulting in the formation of peptide pools.

Peptide Sequence Analysis

Synthesized peptide pools are subjected to sequence analysis to ensure quality and purity. Commonly used methods include high performance liquid chromatography (HPLC) and mass spectrometry. By performing sequence analysis on all peptides (up to 200) in the entire peptide pool, it can be determined that the peptide pool contains all designed peptide sequences. This analysis is applicable to peptide pools used in target screening, antigenic epitope screening, and can further ensure that core sequences are not lost.

Dissolution Testing Services

Customized solubility testing of peptide pools is performed to confirm the appropriate solubility of the mixed peptide pools, with either the default solvent or a customized solvent selected directly.
